Dasatinib for the treatment of gastrointestinal stromal tumors

According to the clinical guidelines and instructions for dasatinib (Dasatinib), dasatinib cannot currently be used to treat gastrointestinal stromal tumors (GIST). Research shows that gastrointestinal stromal tumors begin in "pacemaker" cells in the wall of the gastrointestinal tract, called interstitial cells of Cajal (ICCs), which send signals to the gastrointestinal tract to help food and liquid move through the digestive system. In clinical practice, there is a single-arm, multicenter, Phase 2 trial enrolling patients 18 years and older with advanced GIST who had previously been treated with imatinib and sunitinib. Participants received dasatinib 50 mg orally twice daily for 2 weeks. If tolerated, patients received dasatinib 70 mg twice daily to control tumor progression or unacceptable toxicity. The trial results showed that 2 patients had partial remission, and the disease control rate was 62.0%. Median PFS was 3.0 months. Patients with wild-type GIST had a trend toward longer PFS of 5.5 months. The most commonly observed grade 3 adverse events included anemia and diarrhea. Conclusions: Dasatinib is an effective treatment for patients with gastrointestinal stromal tumors who are refractory to imatinib and sunitinib.
