1. Indications and Usage
Dasatinibis a kinase inhibitor used in treatment
•Newly diagnosed Philadelphia chromosome-positive(Ph+)Chronic myelogenous leukemia(CML)Chronic phase adults.
•Adults with chronic, accelerated, or myeloid or lymphoblastic phasePh+ CML who are resistant or intolerant to prior therapy(including imatinib).
•Adults with Philadelphia chromosome-positive acute lymphoblastic leukemia(Ph+ ALL) who are resistant or intolerant to previous treatments.
•Chronic phasePh+ CMLChildren 1 years old and above.
•Children 1 years old and above who are newly diagnosed with Ph+ ALL and receive chemotherapy.
•Adult chronic phase chronic capsules: 100mg, once daily.
•Adult accelerated phaseCML, myeloid or lymphoblastic phase CML or Ph+ALL: 140 mg once daily.
•Pediatric Chronic Phase Chronic Myeloid Leukemia and Acute Lymphoblastic Leukemia:Starting dose is based on body weight.
•Take orally, with or without food. Do not crush, cut, or chew tablets.
Dosage form and strength
Tablets: 20mg, 50mg, 70mg, pan>80mg, 100mg and 140mg.

Three. Warnings and Precautions
•Myelosuppression and Hemorrhagic Events:Severe thrombocytopenia, neutropenia, and anemia may occur. Use caution if used concurrently with drugs or anticoagulants that inhibit platelet function. Monitor complete blood counts regularly. If indicated, transfuse and discontinue dasatinib.
•Fluid Retention:Fluid retention, sometimes severe, including pleural effusion. Manage with supportive care measures or dose adjustments.
•Cardiovascular Toxicity:Monitor patients for signs or symptoms and initiate appropriate treatment.
•Pulmonary arterial hypertension(PAH):Dasatinib may increase the risk of developing PAH that is reversible when the drug is discontinued. Consider baseline risks and assess patients for PAH during treatmentSigns and symptoms. IfPAH is confirmed, discontinuedasatinib.
•QTProlongation: Use dasatinib with caution in patients who have or may have QT interval prolongation.
•Serious Skin Reactions:Isolated cases of serious mucocutaneous dermatological reactions have been reported.
•Tumor lysis syndrome:Tumor lysis syndrome has been reported. Maintain adequate hydration and correct uric acid levels before initiating treatment with dasatinib.
•Embryo-Fetotoxicity:Can cause harm to the fetus. Inform patients of the potential reproductive risks to the fetus and use effective contraception.
•Effects on Growth and Development in Pediatric Patients:Delayed epiphyseal fusion, osteopenia, growth retardation, and gynecomastia have been reported. Monitor skeletal growth and development in pediatric patients.
•Hepatotoxicity:Assess liver function before initiating treatment and monthly thereafter or as clinically indicated. Monitor hepatic function when used concomitantly with chemotherapy known to be associated with hepatic dysfunction.
Four. Adverse reactions
The most common adverse reactions (≥15%) in patients receiving dasatinib monotherapy included myelosuppression, fluid retention events, diarrhea, headache, rash, bleeding, dyspnea, fatigue, nausea, and musculoskeletal pain.
The most common adverse reactions in pediatric patients receivingdasatinib with combination chemotherapy (≥30%) include mucositis, febrile neutropenia, fever, diarrhea, nausea, vomiting, musculoskeletal pain, abdominal pain, cough, headache, rash, fatigue, constipation, cardiac arrhythmia, hypertension, edema, infection(Bacteria, viruses and fungi), hypotension, decreased appetite, hypersensitivity reactions, dyspnea, epistaxis, peripheral neuropathy and altered state of consciousness.
Five. Drug interactions
•StrongCYP3A4Inhibitors:Dose reduction may be required.
•StrongCYP3A4Inducers:An increase in dose may be required.
•Antacids:Avoid coadministration.
•H2Antagonists and proton pump inhibitors:Avoid coadministration.
6. Use among specific groups of people
•Breastfeeding:It is recommended that women not breastfeed.
