Lorlatinib is a first-generation targeted drug
Lorlatinib is a third-generation tyrosine kinase inhibitor, a new type of targeted drug, mainly used to treat non-small cell lung cancer (NSCLC). As a third-generation targeted drug, lorlatinib has shown excellent efficacy in targeting cancer patients with ALK (tyrosine kinase) and ROS1 (ROS1 kinase) mutations.
The first and second generationALK inhibitors have significant therapeutic effects on some patients, but over time, some patients will develop drug resistance, limiting their long-term efficacy. To address this challenge, third-generation ALK inhibitors such as lorlatinib emerged. Compared with the first two generations, third-generation targeted drugs have a broader inhibitory effect and can deal with a wider variety of ALK and ROS1 mutations, including those that confer resistance to the first two generations of drugs.

The design concept of lorlatinib is to overcome drug resistance and improve the inhibitory effect of the drug on tumor cells. It has demonstrated excellent efficacy in clinical trials, with significant survival advantages especially for patients whose disease has recurred after receiving other treatment modalities.
In addition, the advantage of third-generation targeted drugs lies in their relatively good tolerability and mild side effects, providing patients with a more comfortable treatment experience. However, patients still need to be closely monitored for possible adverse reactions when using lorlatinib to ensure its safety during treatment.
Lorlatinib has been launched in China and is included in medical insurance. Patients can purchase it domestically, but the price is relatively high, about 10,000 to 20,000 yuan. For details, please consult the local hospital pharmacy. There are original lorlatinib drugs and cheaper generic drugs abroad. The cheaper ones are the original drug from Turkey, which costs more than 1,000 yuan, and the original drug from Hong Kong, China, which costs about 30,000 yuan. Generic drugs are mainly generic drugs from Laos and Bangladesh. The price is about several thousand yuan, and the ingredients of the original drugs and generic drugs are basically the same.
