What is the target of larotrectinib?
Larotrectinib is a targeted therapy drug whose main function is to inhibit cancers carrying NTRK (neuronal growth factor receptor kinase) gene fusions. NTRKGene fusion is a rare genetic abnormality found in a variety of cancers, including soft tissue sarcoma, infantile fibroma, lung cancer, etc.
NTRK gene fusion leads to abnormal activity of neuronal growth factor receptor kinase and promotes the growth, differentiation and survival of tumor cells. The design concept of larotrectinib is to precisely inhibit the activity of NTRK kinase, thereby blocking abnormal signaling pathways and inhibiting the development of cancers carrying this genetic abnormality.

Specifically, larotrectinib blocks the activity of the kinase by binding toNTRKkinase, thus inhibiting the transmission of downstream signaling pathways. This highly targeted mechanism of action allows the drug to precisely intervene in the abnormal biological activity caused by NTRK gene fusion, thereby preventing the growth and spread of cancer cells.
Importantly, larotrectinib's targeted treatment strategy provides patients with a personalized treatment option. Before receiving treatment, patients often undergo genetic testing to confirm whether their tumors carry NTRK gene fusions and thus determine whether they are candidates for treatment with larotrectinib.
Larotrectinib has been launched in China and has not yet been included in medical insurance. Since it has not been launched in China for a long time, it may be difficult to purchase it in hospitals. For specific conditions, it is recommended to consult the local hospital pharmacy. In foreign markets, larotinib has two options: original drug and generic drug. Original drugs include the German version, the American version, the European version and the Hong Kong version, and the price is about 10,000 to 20,000 yuan. In comparison, generic drugs are much cheaper. There are Lao and Bangladeshi versions of generic drugs, and the price is around two to three thousand. It is worth noting that the ingredients of original drugs and generic drugs are basically the same.
