Can Ivonib treat chondrosarcoma?
Surgery is the mainstay of treatment for localized chondrosarcoma; there are no known effective systemic therapies for locally advanced and/or metastatic disease. Up to 65% of chondrosarcomas have isocitrate dehydrogenase 1/2 (IDH1/2) enzyme mutations, leading to the accumulation of the tumor metabolite D-2-hydroxyglutarate (2-HG). Ivosidenib (Ivosidenib) is a selective inhibitor of mutated IDH1 approved in the United States for specific cases of acute myeloid leukemia.

In a Phase I study, patients with chondrosarcoma tolerated Ivosidenib well, and Ivosidenib resulted in significant reductions in plasma and tumor 2-HG, indicating that the drug's efficacy was on target. The disease stable rate was 52%, the median progression-free survival (PFS) was 5.6 months, and the 6-month PFS rate was 39.5%. Furthermore, the higher PFS rate in patients without dedifferentiated histology suggests that ivonib may be more effective against conventional chondrosarcomas. The incidence of treatment-related grade 3 adverse reactions was low, and some patients reported adverse events related to prolongation of the QT interval on the electrocardiogram.
The original version of ivonib has been launched in China, but it is not covered by medical insurance. The price of domestically sold specifications0.25g*60 tablets per box is as high as 100,000 yuan, while the price of the original American version sold overseas is as high as more than 200,000 yuan per box (the price may fluctuate due to exchange rates). Generic drugs of Ivonib are also sold overseas. Their drug ingredients are basically the same as those of the original drugs sold domestically and abroad. For example, the price of 250mg*60 tablets produced by a Laos pharmaceutical factory may be more than 6,000 yuan per box (the price may fluctuate due to the exchange rate), and the price is relatively cheap.
