Medication Instructions for Patients: How to Determine the Dosage of Eltrombopag/Eltrombopag

Eltrombopag/Eltrombopag is a drug that stimulates the body to produce platelets, which is essential for maintaining normal blood clotting function. It is commonly used to treat patients with immune thrombocytopenia (ITP) and some aplastic anemias. This medication is available in various dosage forms, including 12.5 mg, 25 mg, 50 mg, and 75 mg tablets.

For patients with persistent or chronic ITPthe dosage needs to be adjusted according to the patient's specific conditions. Pediatric patients aged 1 to 5 years are recommended to take 25 mg daily (i.e. one 25 mg tablet). For adults and children with ITP aged 6 years and older, the recommended daily dose is 50 mg (one 50 mg tablet). For patients with ITP and mild, moderate, or severe hepatic impairment (Child-Pugh classes A, B, C), the initial dose is 25 mg daily (one 25 mg tablet). ITP patients of East Asian/Southeast Asian ancestry are also recommended to start treatment with a daily dose of 25 mg. For these patients who also suffer from liver impairment, the initial dose can even be reduced to 12.5 mg per day (one 12.5 mg tablet).

For first-line treatment of severe aplastic anemia, the medication regimen is different. For children aged 2 to 5 years old, while receiving standard immunosuppressive treatment, the daily dose needs to be calculated based on body weight. The recommended dose is 2.5 mg per kilogram of body weight. For children aged 6 to 11 years, the recommended daily dose is 75 mg (one 75 mg tablet). For patients 12 years and older, the daily dose is 150 mg (two 75 mg tablets or other equivalent combination) for 6 months. For patients with severe AA of East/Southeast Asian ancestry or those with hepatic impairment, the first-line treatment dose should be reduced by 50%.

For the treatment of refractory severe aplastic anemia, the recommended initial dose is 50 mg daily (one 50 mg tablet). Patients of East/Southeast Asian ancestry or those with hepatic impairment should start with a daily dose of 25 mg. After that, the dose can be increased by 50 mg every two weeks according to the situation until the platelet count reaches or exceeds 50×109/L. But please note that the maximum daily dose should not exceed 150 mg.
