Can selumetinib completely cure café-au-lait spots?

Café freckles are a common skin disease that usually begins at birth and appears as light brown spots on the skin that resemble the color of coffee solution with milk added, hence the name. Café-au-lait spots are usually benign and do not require treatment, but for people with numerous or highly visible café-au-lait spots, they can cause serious psychological and social problems.

Selumetinib (Selumetinib) is a targeted therapy drug commonly used to treat some rare genetic diseases, such as neurofibromatosis. However, its use in café au lait treatment is not currently standard practice, and there is insufficient evidence that it can cure café au lait.

Café-au-lait spots appear in patients with neurofibromatosis because neurofibromatosis type 1 (NF1; OMIM 16220) is caused by loss of function caused by pathogenic variants in NF1. The oral MEKinhibitor selumetinib can reduce NF1 the volume of plexiform neurofibromas (PN) in children NF1 and relieve café-au-lait spots. Selumetinib is not a drug for treating simple café-au-lait spots.

There is currently no treatment that can completely cure café-au-lait spots, but some research suggests selumetinib may be helpful in reducing the color and size of café-au-lait spots. Selumetinib, as a signal transduction inhibitor, affects cell proliferation and differentiation by inhibiting specific signaling pathways, which may affect the formation and development of café-au-lait spots.

However, it should be pointed out that selumetinib is not currently officially approved for the treatment of café-au-lait spots, and its efficacy and safety in this field still need to be confirmed by more clinical studies. In addition, drug treatment may be accompanied by a series of side effects and risks. Therefore, when considering using selumetinib to treat café-au-lait spots, a full risk assessment must be conducted and treatment under the guidance of a doctor.
