Is neratinib/neratinib effective after dual targeting of breast cancer?
Neratinib/Neratinib is a tyrosine kinase inhibitor (TKI) in the form of an oral drug. It plays an important role in the treatment of breast cancer, especially the treatment of HER2-positive breast cancer. For breast cancer patients who have undergone dual-targeted therapy, neratinib may be a treatment option worth considering. However, whether neratinib is suitable for use still needs to be evaluated by a doctor based on the patient's specific situation.

As an irreversible panHER inhibitor, the therapeutic effect of neratinib surpasses other similar drugs, such as lapatinib, in many aspects. It not only enhances trastuzumab-mediated cytotoxicity (ADCC) but also induces cell cycle arrest and proliferation inhibition in HER2-expressing cells. This gives neratinib a significant advantage in inhibiting tumor growth and spread.
In early clinical trials, neratinib showed an efficacy rate of up to 73% in patients with HER2-positive metastatic breast cancer who had previously received adjuvant or neoadjuvant trastuzumab treatment. However, at the same time, it is also accompanied by certain toxic reactions, such as diarrhea, which affects the patient's tolerance and treatment compliance to a certain extent. Neratinib still showed encouraging results when tested in patients with early-stage HER2-positive breast cancer. It significantly improves patients' disease-free survival and provides new treatment hope for patients with early-stage breast cancer.
Currently, neratinib is undergoing clinical trials in combination with other therapies such asT-DM1 and fulvestrant. In studies targeting brain metastases, as shown in trials such as NEfERT-T, NALA and TBCRC 022, neratinib has shown promise in reducing the incidence of central nervous system events. These clinical trial results provide strong support for the application of neratinib.
