Tucatinib, also known as tucatinib or tucatinib, is a potent kinase inhibitor widely used in the combination treatment of HER2-positive breast cancer and RAS wild-type HER2-positive colorectal cancer. Its chemical name is (N4-([1,2,4]triazolo[1,5-a]pyridin-7-oxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrazol-2-yl)quinazoline-4,6-diamine. On the market, tucatinib is available as oral film-coated tablets in two dosages: 50 mg and 150 mg, and its trade name is widely recognized as TUKYSA. The drug has shown remarkable efficacy in treating certain cancers. Specifically, it can be used in combination with trastuzumab and capecitabine to treat adult patients with advanced unresectable or metastatic HER2-positive breast cancer, especially those who have experienced one or more anti-HER2 treatment regimens, including patients with brain metastases. In addition, tucatinib in combination with trastuzumab is an effective treatment option for adults with RAS wild-type HER2-positive unresectable or metastatic colorectal cancer whose disease has progressed despite fluoropyrimidine-, oxaliplatin-, and irinotecan-based chemotherapy.

As a HER2 tyrosine kinase inhibitor, tucatinib works by effectively inhibiting the growth mechanism of HER2-expressing tumors. Studies have shown that the combination of tucatinib and trastuzumab significantly increases anti-tumor activity compared to use alone.
