Nilotinib/nilotinib usage and dosage

Nilotinib (Nilotinib) is an important drug mainly used to treat adult patients with chronic phase or accelerated phase of Philadelphia chromosome-positive chronic myelogenous leukemia (Ph+ CML) who are resistant or intolerant to previous treatments (including imatinib).

For adult patients, the recommended dose of nilotinib is 400mg twice daily with an interval of approximately 12 hours between doses. Such dosage and frequency help maintain the effective concentration of the drug in the body, thereby achieving the best therapeutic effect.

Nilotinib should be taken with food to ensure full absorption of the drug. It is recommended to take it at least 1 hours before meals or at least 2 hours after meals. This can avoid the impact of food on drug absorption and ensure that the drug can quickly and effectively enter the blood circulation. Nilotinib should be swallowed whole in capsule form and should not be chewed, sucked, or opened. This helps protect the medication from damage by stomach acid and digestive enzymes, ensuring stable release of the medication in the intestines.

For patients who are unable to swallow capsules, the capsule contents can be mixed with a teaspoon of applesauce and taken immediately. However, it should be noted that the amount of applesauce cannot exceed one teaspoon and cannot be eaten at the same time as other foods to avoid affecting the efficacy of the medicine.

If the electrocardiogram showsQTc>480 milliseconds, you should stop taking nilotinib and measure serum potassium and magnesium in a timely manner. Depending on the recovery of QTc and serum potassium and magnesium levels, doctors may adjust the dose or suspend use.

If hematologic toxicity occurs, use of nilotinib should be suspended. After the blood picture returns to normal levels, the drug can be resumed at the initial dose. If the blood count remains low and does not recover, your doctor may consider lowering the dose.
