However, the structure of avatrombopag does not contain a hydrazide structure but a benzothiazole structure, which is not easily chelated with metal cations. In contrast, heltrombopag needs to be taken on an empty stomach to avoid concurrent ingestion with food or medications containing metal ions.

The bioavailability of avatrombopag is as high as100%, which means that the drug is fully absorbed after oral administration. The bioavailability of Hetrombopag is low, only 38%, indicating that some drugs are not absorbed after oral administration. The half-life of avatrombopag is 7-9 hours, and its efficacy is long-lasting. In contrast, the half-life of Hetrombopag is 2-3 hours, and its efficacy is relatively short.

The steady-state plasma concentration of avatrombopag is 1.5-3.0 ng/mL, which can effectively stimulate platelet production. The steady-state plasma concentration of Hetrombopag is relatively low, ranging from 0.3 to 0.8 ng/mL.

Avatrombopag is indicated primarily for the treatment of adult patients with thrombocytopenia associated with chronic liver disease who are scheduled for elective diagnostic procedures or surgery. Hatrombopag is suitable for adult patients with chronic primary immune thrombocytopenia(ITP)who have had poor response to glucocorticoids, immunoglobulins and other treatments due to thrombocytopenia and clinical conditions that increase the risk of bleeding.

Both have shown good efficacy in treating their respective indications, but which one is better depends on the specific situation of the patient and the judgment of the clinician.

Both drugs may produce certain side effects during use, such as headache, nausea, etc. However, since avatrombopag is not affected by food, patient medication compliance may be relatively good.
