What diseases does fostamatinib treat?

Fostamatinib(fostamatinib) is a tyrosine kinase inhibitor with activity against spleen tyrosine kinase (SYK). Fostatinib Specifically indicated for the treatment of thrombocytopenia in adult patients with chronic immune thrombocytopenia who has had an inadequate response to prior therapy (ITP).

The U.S.FDA approval of fostatinib was based on data from the FIT clinical program. In two parallel, phase 3, multicenter, randomized, double-blind, placebo-controlled trials (FIT1 and FIT2), patients with persistent/chronic ITP were randomized 2:1 to fostamatinib (n=101) or placebo (n=49) 100mg BID for 24 weeks, with the dose in non-responders increased to 150mg BID after 4 weeks. The primary endpoint was stable response (weeks 14-24, ≥4 of 6 biweekly visits, platelets ≥50,000/μL, no rescue therapy). The median platelet count at baseline was 16,000/μL; the median duration of ITP was 8.5 years. Stable remission occurred in 18% of patients in the fostamatinib group compared with 2% of patients in the placebo group (P=0.0003). The overall response rate (retrospectively defined as ≥1 platelet count ≥50,000/μL within 12 weeks before treatment) occurred in 43% of patients treated with fostamatinib. and 14% in the placebo group (P=0.0006). Median time to response was 15 days (100 mg daily), with 83% responding within 8 weeks.

Fostatinib has not yet been launched in the domestic market.
