Dasatinib (Dasatinib) is a small molecule targeted drug mainly used to treat certain types of leukemia, especially chronic myelogenous leukemia (CML) and acute lymphoblastic leukemia (ALL). It belongs to the second generation tyrosine kinase inhibitor (TKI), which mainly works by inhibiting the tyrosine kinase activity of BCR-ABL fusion protein. Dasatinib is approved in the United States and the European Union and is active against most clinically relevant BCR-ABL mutations, except for the highly resistant T315I.

Dasatinib has a unique mechanism of action. BCR-ABL fusion protein is produced by the translocation of chromosome 9 and chromosome 22. It is abnormally expressed in leukemia cells, leading to uncontrolled cell proliferation. Dasatinib binds to the BCR-ABL protein and blocks its kinase activity, thereby inhibiting the proliferation of cancer cells and promoting cell apoptosis. This mechanism allows dasatinib to achieve clinical therapeutic effects, especially in patients who are resistant to first-generation TKIs such as imatinib. Dasatinib was approved by the U.S. Food and Drug Administration (FDA) in 2006 and has become an important option for the treatment of CML. Compared with the first-generation TKI, dasatinib has a stronger inhibitory effect, and it also shows good anti-tumor activity against certain mutations (such as T315I mutation). Although dasatinib has achieved good efficacy in clinical application, it is also associated with some adverse reactions.
