What are the side effects of ivonib/ivitinib?

Ivosidenib/Ivosidenib targets a specific gene mutation called IDH1 that affects your bone marrow. IDH1 mutations prevent young blood cells from developing into healthy adult blood cells, which can cause symptoms of acute myeloid leukemia (AML). Ivosidenib is also used to treat AML in adults 75 years and older who have newly diagnosed AML and are unable to receive certain chemotherapy treatments because of other health problems; and to treat cholangiocarcinoma that has progressed or spread to other parts of the body (metastatic) after treatment with other drugs, and myelodysplastic syndrome (MDS).

Avosidenib is only indicated for the treatment of AML in adults with IDH1 mutations and after other treatments have failed or stopped working. Doctors test patients' cancers for this genetic mutation. The most common side effects of ivonib when used in combination with azacitidine to treat AML include vomiting, neutropenia (low levels of neutrophils, a type of infection-fighting white blood cell), thrombocytopenia (low levels of platelets), and prolongation of the QT interval (abnormal activity of the heart that affects heart rhythm). (difficulty falling and staying asleep, and poor sleep quality); when used alone to treat biliary tract cancer, the most common side effects of ivosidenib include fatigue, nausea, abdominal pain, diarrhea, decreased appetite, ascites (fluid accumulation in the abdomen), vomiting, anemia (low levels of red blood cells), and rash.

Avosidenib can cause serious adverse reactions, includingQTc interval prolongation and Guillain-Barre syndrome; Ivosidenib should not be taken with drugs known as "strong CYP3A4 inducers" because these drugs may affect blood levels of Ivosidenib. Additionally, it should not be used with dabigatran (an "anticoagulant" or blood-thinning drug) or in patients with certain heart rhythm problems.
