How does ensidipine work?

Enasidenib (Enasidenib) is a targeted drug mainly used to treat specific types of leukemia, especially relapsed or refractory acute myeloid leukemia (AML) carrying isocitrate dehydrogenase 2 (IDH2) gene mutations.

Ensidipine, as an IDH2 inhibitorinterferes with the growth and division of AML cells by interfering with the abnormal protein produced by IDH2 gene mutations. IDH2 gene mutation is a common mutation in AML patients. The mutation leads to abnormal enzyme activity and the production of large amounts of 2-hydroxyglutarate (2-HG). The accumulation of 2-HG interferes with normal hematopoietic function and further promotes the growth and division of leukemia cells. Ensidipine can inhibit the enzyme activity of the IDH2 mutant gene and reduce the production of 2-HG, thereby restoring normal hematopoietic function.

Ensidipine reduces the proliferation and viability of leukemia cells by inhibiting the activity ofIDH2 mutants. Ensidipine can promote the differentiation of leukemia cells into normal mature blood cells, restore normal hematopoietic function, and help reduce the number of abnormal leukemia cells. Clinical trials have shown that ensidipine can significantly prolong the survival of patients with R/R AML, and complete remission can be observed in a subset of patients. By controlling the progression of the disease, ensidipine may alleviate the symptoms of anemia, bleeding, infection and other symptoms commonly seen in patients with leukemia, and improve the quality of life of patients.
