How effective is Revuforj (Revumenib)?

Revuforj (Revumenib) is the first menin-like drug to be administered orally for patients with KMT2A translocation acute leukemia. KMT2A translocation involves a structural change in the lysine transferase 2A gene, causing part of the chromosome to break and attach to another chromosome, causing an aggressive form of cancer. Treatment of this condition has always presented significant challenges because the prognosis is often poor.

Revuforj inhibits the interaction between KMT2A and menin protein, thereby changing the transcription pattern of multiple genes, thereby affecting the growth and behavior of cancer cells. This mechanism gives Revuforj a unique advantage in the treatment of KMT2A translocation-related acute leukemia.

In theAUGMENT-101 clinical trial, researchers evaluated 104 patients with relapsed or refractory KMT2A translocation acute leukemia. The results showed that 21% of patients achieved complete remission, and the average duration of complete remission in these patients reached 6.4 months. Additionally, 23% of patients who received Revuforj went on to receive a stem cell transplant. These data show a significant improvement compared with the previous complete response rate of 5%, reflecting the potential clinical value of Revuforj in this type of high-risk patients.

The approval of Revuforj by the U.S. Food and Drug Administration (FDA) marks a major breakthrough in the treatment of relapsed/refractory acute leukemia associated with KMT2A translocation. Such genetic changes are closely associated with poor prognosis, making the clinical application of this new drug of great significance.
