In addition, defibrinated sodium can promote the release of prostaglandin I2 (PGI2) and prostaglandin E2 (PGE2), thereby causing vasodilation, inhibiting platelet aggregation, and reducing ischemic damage. Studies have shown that defibrinated sodium can significantly increase the expression of thrombin regulatory protein (TM) and tissue factor pathway inhibitors to produce anticoagulant effects.

Defibrotide sodium is an injectable drug and the recommended dose is 6.25 mg/kg given every 6 hours as a 2-hour intravenous infusion. Treatment lasts for a minimum of 21 days. If the signs and symptoms of VOD do not resolve after 21 days, continue treatment until resolution. Hypersensitivity reactions have occurred in less than 2% of patients treated with defibrotide. If a severe hypersensitivity reaction occurs, discontinue defibrinated sodium and follow standard medical care and monitor until symptoms resolve.

(Defibrotide) is the first drug approved by the US FDA to treat severe hepatic veno-occlusive disease, providing a new treatment option for this rare disease and bringing new treatment hope.
