Defibrotide sodium can be administered via intravenous injection or oral administration. In view of factors such as hydrolytic inactivation of digestive tract DNase, small intestinal absorption efficiency, and liver first-pass effect, the oral bioavailability of defibrinated sodium is only 58% to 70%. Acute toxicity experiments have been carried out on mice, rats, rabbits and dogs, and no toxic reactions occurred in the experimental animals. Two generations of reproductive toxicity and teratogenic experiments confirmed that it has no reproductive toxicity to rats and rabbits. Defitelio has been proven to have no immunotoxicity. No allergic reactions occurred in the test animals, and it did not affect their cellular immune responses. Experimental results on chromosomal aberrations and gene mutations in mammalian cells show that defibrinated sodium has no mutagenic risk and can antagonize the mutagenic activity of certain chemical carcinogens (dimethylbenzanthracene, daunorubicin, and benzopyrene).
