Carcinogenicity studies of oral administration of a racemic mixture (omeprazole) in rats identified gastric enterochromaffin-like (ECL) cell hyperplasia and carcinoid tumors. These effects are secondary to the sustained reduction in gastric acid production and the marked hypergastrinemia seen in rats after chronic administration of gastric acid secretion inhibitors. Using any method, including proton pump inhibitors, to reduce the acidity of gastric juice will increase the number of bacteria in the stomach (usually found in the gastrointestinal tract). Treatment with proton pump inhibitors may result in a slightly increased risk of gastrointestinal infections (such as Salmonella and Campylobacter).
The safety and effectiveness of esomeprazole treatment have been verified in two trials: a clinical comparative study of esomeprazole and omeprazole in the treatment of reflux esophagitis. The total effective rate of esomeprazole was 97.18%, which was significantly higher than that of omeprazole, which was 84.51%.
